Overview

A mitochondrial oxidative phosphorylation uncoupler is a molecule that disrupts the normal coupling between the electron transport chain (ETC) and ATP synthesis in mitochondria, leading to increased oxygen consumption and heat production, while reducing or halting ATP synthesis. Uncouplers act primarily by transporting protons across biological membranes, bypassing ATP synthase, ultimately reducing efficiency of oxidative phosphorylation. Endogenous uncoupling proteins also mediate regulated physiological uncoupling for thermogenesis or protection against reactive oxygen species.
