Overview

Mitochondrial protein thiols represent the collective pool of sulfhydryl (-SH) groups located on cysteine residues within mitochondrial proteins. These thiols are critical components of the mitochondrial redox sensing and signaling apparatus, serving as primary targets for reactive oxygen and nitrogen species (ROS/RNS). The oxidation state of these thiols regulates the activity of numerous enzymes involved in the Krebs cycle, oxidative phosphorylation, and the mitochondrial permeability transition pore (mPTP). In many pathological conditions, such as Parkinson's disease and myocardial infarction, excessive oxidative stress leads to the depletion or irreversible oxidation of these thiols, resulting in mitochondrial dysfunction and cell death. Therapeutic strategies often involve the use of mitochondria-targeted antioxidants or thiol-restoring agents to maintain the integrity of the mitochondrial thiol proteome and preserve cellular bioenergetics.

Mechanism of action
