Overview

The mitochondrial quality control pathway is a collective term for a series of interconnected cellular pathways that monitor and maintain mitochondrial function and integrity. These include mechanisms for detecting and repairing mitochondrial damage, removing dysfunctional mitochondrial components via mitophagy, regulating mitochondrial dynamics (fusion and fission), promoting mitochondrial biogenesis, and maintaining proteostasis through chaperones and proteases. Failure or dysfunction in these pathways is implicated in numerous diseases, such as neurodegenerative disorders, cardiovascular diseases, cancer, metabolic syndromes, and inflammatory conditions. Therapeutic interventions aim to enhance or normalize these MQC strategies to protect against disease and cellular dysfunction, but targeting the pathway poses challenges due to its complexity and the central role of mitochondria in basic cell biology.
