Overview

Mitochondrial ribosomal protein L14 (MRPL14) is a nuclear-encoded protein that is an essential component of the 39S large subunit of the mitochondrial ribosome. It plays a structural role in the mitochondrial translation machinery, which synthesizes 13 essential polypeptides encoded by mitochondrial DNA, all of which are required for assembly and function of the oxidative phosphorylation (OXPHOS) complexes. MRPL14 interacts directly with other mitoribosome assembly factors, such as C7orf30, which is required for efficient incorporation of MRPL14 into the large subunit. Depletion of MRPL14 severely impairs mitochondrial ribosome biogenesis, resulting in diminished mitochondrial translation and compromised formation of respiratory chain complexes. There are no current drugs or therapeutics that target MRPL14, and it is not considered a classical therapeutic target such as a receptor, enzyme, or transporter[1][2][5].
