Mitochondrial ribosomal protein L46 (MRPL46) is a nuclear-encoded protein that functions as a structural constituent of the 39S large subunit of the mitochondrial ribosome (mitoribosome)[1][2][4]. Its primary role is to contribute to the assembly and function of the mitoribosome, which synthesizes proteins within mitochondria that are crucial for mitochondrial respiratory chain complexes and cellular energy production[1][4][5][6]. MRPL46 participates in maintaining translation fidelity and proper mitoribosome assembly[9]. Mitochondrial ribosomal proteins such as MRPL46 exhibit unique non-redundant roles in diverse tissues, especially those with high energy demands[3][4]. Mutations or dysregulation of MRPL46 and other MRPs have been linked to various mitochondrial disorders and some cancers[2][3][4][11]. No known small molecule drugs or direct modulators target MRPL46, and it is not currently considered a therapeutic target but rather a critical housekeeping protein essential for mitochondrial protein synthesis and cellular metabolism[1][2][4][9][12].
