Overview

Mitochondrial ribosomal protein L50 (MRPL50) is a nuclear-encoded protein and a component of the 39S large subunit of the mammalian mitochondrial ribosome (mitoribosome)[1][4][6]. Mitoribosomes are essential for protein synthesis within mitochondria, specifically synthesizing key components required for oxidative phosphorylation and cellular energy production[1][2][3]. Each ribosomal protein in the mitochondrial ribosome, including MRPL50, plays a unique and essential role, with minimal redundancy among family members, as demonstrated by mouse knockout studies showing early embryonic lethality when individual MRPs are missing[5]. MRPL50 is widely and ubiquitously expressed in tissues, consistent with its fundamental role in cellular viability and energy production[5]. Defects or mutations in MRPL50 and other MRPs have been linked to mitochondrial disorders such as combined oxidative phosphorylation deficiency and Leigh disease, and increasingly, to roles in cancer due to the involvement of mitoribosomes in cell proliferation and apoptosis[1][5]. However, to date, MRPL50 itself is not recognized as a direct therapeutic target, receptor, enzyme, or transporter, and no drugs are known to specifically interact with or modulate this protein.
