Mitochondrial ribosomal protein S28 (MRPS28) is a nuclear-encoded protein that functions as a structural component of the small 28S subunit of the mitochondrial ribosome (mitoribosome). It is essential for mitochondrial protein synthesis, facilitating the translation of mitochondrial DNA-encoded proteins involved in oxidative phosphorylation. The mitoribosome differs from prokaryotic ribosomes in protein to rRNA ratio and composition. Mutations in the MRPS28 gene have been associated with multisystem mitochondrial disorders, such as combined oxidative phosphorylation deficiency 47 and abdominal obesity-metabolic syndrome 1, but MRPS28 itself is not a common direct therapeutic target.
