Overview

Mitogen-activated protein kinase–activated protein kinase 5 (MAPKAPK5), also known as PRAK, is a serine/threonine-protein kinase and a member of the MAPKAP kinase subfamily. It acts as a tumor suppressor serine/threonine-protein kinase involved in mTORC1 signaling and post-transcriptional regulation. It is activated by cellular stress and proinflammatory cytokines and phosphorylates several substrates including heat shock protein HSP27, FOXO3, ERK3/MAPK6, and ERK4/MAPK4. It plays roles in cellular stress response pathways and regulation of cell motility. Loss or reduced activity may contribute to cancer progression, and reduced levels have been linked with Alzheimer's disease.
