Overview

Mitogen-activated protein kinases (MAPKs) are a family of serine/threonine protein kinases that transduce extracellular signals into cellular responses. MAPK1 (commonly known as ERK2), MAPK8 (JNK1), and MAPK14 (p38α) are prototypic MAPK subfamily members, each activated by distinct upstream kinases and mediating specific, sometimes overlapping biological processes. MAPK1 is majorly involved in growth factor-mediated signaling and cell cycle progression. MAPK8 mediates responses to stress and is strongly implicated in apoptosis. MAPK14 is critical for inflammatory signaling, cellular responses to environmental stress, and is often targeted in immune and inflammatory diseases. While each can be targeted therapeutically, their distinct biology and druggability profiles necessitate careful target selection and validation in therapeutic settings[1][2][4][5]. Note: MAPK1, MAPK8, and MAPK14 are each separate, well-defined kinase targets; listing them together is incorrect unless specifically referring to them as a functional group (e.g., MAPK family).
