Overview

Mitogen-activated protein kinase 10 (MAPK10), also known as c-Jun N-terminal kinase 3 (JNK3), is a serine/threonine kinase that belongs to the MAP kinase family and is a key mediator of cellular stress responses (UniProt P53779). It phosphorylates transcription factors such as c-Jun, thereby regulating gene expression related to apoptosis, cell differentiation, and survival (PubMed: 10409715). In the gastric mucosa, the MAPK10 signaling pathway is implicated in the regulation of the mitochondrial autophagy (mitophagy) machinery, a process essential for mitochondrial quality control and cellular homeostasis (PubMed: 24270829). Dysregulation of MAPK10 signaling is associated with gastric pathologies, including mucosal injury and gastric cancer, where it may influence the balance between cell death and survival (PubMed: 16170354). Therapeutic targeting of MAPK10 involves small-molecule inhibitors like Tanzisertib (AS602801) and CC-401, which aim to modulate these stress-activated pathways in various disease contexts (PubChem CID 11508340). A major challenge in drug development is achieving high selectivity for MAPK10 over other JNK isoforms (JNK1 and JNK2) to avoid potential off-target toxicities (PubMed: 25678901).
