Overview

Extracellular-signal-regulated kinase 1 (ERK1), also known as MAPK3, is a serine/threonine protein kinase and a key component of the mitogen-activated protein kinase (MAPK) signaling pathway. It mediates signal transduction from the cell surface to the nucleus in response to various extracellular stimuli, playing a role in cell proliferation, differentiation, adhesion, migration, and survival. ERK1 is activated by MEK1/2 via phosphorylation at Thr202/Tyr204. Dysregulation of the Ras-Raf-MEK-ERK pathway is implicated in cancer.

Mechanism of action

ERK1 is phosphorylated and activated by MEK1/2, leading to downstream phosphorylation of numerous substrates, including transcription factors and regulatory proteins. Inhibitors targeting upstream kinases (e.g., BRAF) indirectly affect ERK1 activity.
