Overview

Mitogen-activated protein kinase 6 is an atypical serine/threonine protein kinase belonging to the MAPK (mitogen-activated protein kinase) family, encoded by the MAPK6 gene in humans. Unlike classical MAPKs, MAPK6 (also known as ERK3) features distinct structural and regulatory properties, including a unique phosphorylation motif and extended regulatory regions. MAPK6 is primarily located in the nucleus and is involved in the integration of multiple biochemical signaling pathways in response to external stimuli. Recent insights from crystal structure analysis highlight its distinct ATP-binding pocket, creating opportunities for selective drug development. Although the full spectrum of its biological roles remains under investigation, MAPK6 plays a part in cell migration and has been implicated in the invasiveness of certain cancers. It is currently considered a promising, but understudied, therapeutic target for inhibitor-based cancer therapy.
