Overview

Mitogen-activated protein kinase 7 (MAPK7) mRNA, commonly known as ERK5 mRNA, encodes the Extracellular Signal-Regulated Kinase 5 protein, a distinct member of the MAP kinase family (UniProt: Q13164). ERK5 is unique among MAPKs due to its large size and a C-terminal transcriptional activation domain that allows it to function as both a kinase and a transcriptional co-activator (PubMed: 26071358). The mRNA transcript is a primary target for gene-silencing approaches, such as small interfering RNAs (siRNAs) and antisense oligonucleotides (ASOs), which are designed to reduce ERK5 protein levels in pathological conditions (PubMed: 30610119). ERK5 signaling is frequently upregulated in various malignancies, including breast, prostate, and colorectal cancers, where it drives cell proliferation, survival, and the epithelial-to-mesenchymal transition (PubMed: 22508478). Beyond oncology, ERK5 mRNA expression is critical in cardiovascular health, particularly in maintaining endothelial cell function and vascular integrity (PubMed: 25659601). Targeting the mRNA provides a therapeutic alternative to small-molecule inhibitors, potentially offering higher specificity by preventing the synthesis of the ERK5 protein entirely.
