Mitogen-activated protein kinase 8-interacting protein 2 (MAPK8IP2) is a cytoplasmic scaffold protein that aggregates and organizes key proteins in the c-Jun N-terminal kinase (JNK) signaling pathway[2][3][7][8]. Highly expressed in the brain and pancreatic beta cells, MAPK8IP2 regulates the activity of JNK1 (MAPK8) and MKK7 (MAP2K7) kinases and modulates synaptic transmission by influencing the ratio of AMPA to NMDA glutamate receptors at synapses[2][3]. This protein is also implicated in anti-apoptotic signaling in insulin-secreting cells and in vesicular transport through its interactions with motor proteins[8]. Loss of MAPK8IP2 is associated with Phelan-McDermid syndrome, featuring intellectual disability and other neurological deficits[2][3]. The protein is a member of the JIP family of scaffold proteins and participates in multiple signaling pathways, especially those influencing cell death, neurotransmission, and possibly some cancers[3][7].
