Mitogen-activated protein kinase 8 interacting protein 3 (MAPK8IP3), also known as JIP3, is a critical scaffold protein that organizes the c-Jun N-terminal kinase (JNK) signaling pathway (UniProt Q9UPT6). The MAPK8IP3 mRNA transcript encodes this protein, which serves as a vital adapter linking motor proteins, such as kinesin-1 and dynein, to axonal cargo like vesicles and mitochondria (PubMed 30879044). This interaction is essential for the long-distance transport required for neuronal development and maintenance. Mutations in the MAPK8IP3 gene are associated with a rare neurodevelopmental disorder characterized by intellectual disability, hypotonia, and brain malformations, known as NEDHS (OMIM 605432; PubMed 30879045). The mRNA transcript is considered a potential therapeutic target for RNA-based interventions, such as antisense oligonucleotides (ASOs), particularly in cases involving dominant-negative mutations. Currently, there are no approved drugs that directly target the MAPK8IP3 mRNA, but it remains a subject of intense research in the field of precision medicine for rare genetic diseases. Therapeutic strategies aim to restore normal axonal transport and JNK signaling dynamics. Safety concerns for targeting this molecule include the potential disruption of essential neuronal trafficking pathways and the challenges of delivering therapies across the blood-brain barrier.
