Mitogen-activated protein kinase 8 interacting protein 3 (MAPK8IP3), also known as JIP3, is a critical scaffold protein that organizes the c-Jun N-terminal kinase (JNK) signaling pathway (UniProt Q9UPT6). The MAPK8IP3 mRNA transcript encodes this protein, which serves as a vital adapter linking motor proteins, such as kinesin-1 and dynein, to axonal cargo like vesicles and mitochondria (PubMed 30879044). This interaction is essential for the long-distance transport required for neuronal development and maintenance. Mutations in the MAPK8IP3 gene are associated with a rare neurodevelopmental disorder characterized by intellectual disability, hypotonia, and brain malformations, known as NEDHS (OMIM 605432; PubMed 30879045). The mRNA transcript is considered a potential therapeutic target for RNA-based interventions, such as antisense oligonucleotides (ASOs), particularly in cases involving dominant-negative mutations. Currently, there are no approved drugs that directly target the MAPK8IP3 mRNA, but it remains a subject of intense research in the field of precision medicine for rare genetic diseases. Therapeutic strategies aim to restore normal axonal transport and JNK signaling dynamics.
