Overview

Mitogen-activated protein kinase JNK (MAPK JNK), also known as c-Jun N-terminal kinase (JNK), is a subfamily of the mitogen-activated protein kinases (MAPKs). These kinases are serine/threonine-specific enzymes that play crucial roles in transducing extracellular signals into various intracellular responses, particularly those related to stress and inflammation. The JNK family consists of three main isoforms encoded by separate genes: JNK1 (MAPK8), JNK2 (MAPK9), and JNK3 (MAPK10). Activation occurs via dual phosphorylation on threonine and tyrosine residues within a Thr-Pro-Tyr motif by upstream kinases MKK4 and MKK7. JNKs regulate several key cellular processes, including transcriptional regulation, cell proliferation, differentiation, apoptosis, and inflammatory responses. Dysregulation of the MAPK/JNK pathway has been implicated in neurodegenerative diseases, inflammatory conditions, and infectious diseases.
