Overview

MEK1 and MEK2 are closely related dual-specificity protein kinases that play a central role in the RAS-RAF-MEK-ERK signal transduction cascade. This pathway is crucial for regulating diverse cellular processes such as apoptosis, cell cycle progression, migration, differentiation, metabolism, and proliferation. MEK1/2 phosphorylate and activate ERKs (extracellular signal-regulated kinases), specifically ERK1/ERK2. The pathway is frequently hyperactivated due to upstream mutations in RAS or BRAF genes—a common event across many human cancers—making MEKs key therapeutic targets.
