Overview

Mitogen-activated protein kinase kinase kinase (MAP3K) refers to a family of serine/threonine protein kinases functioning as critical upstream activators in the MAPK signaling pathway. They receive signals from extracellular mitogens, stress factors, and cytokines and propagate these signals by phosphorylating MAP kinase kinases (MAP2K), which in turn activate MAP kinases responsible for diverse cellular responses such as proliferation, apoptosis, differentiation, and immune function. Specific MAP3Ks, such as the RAF family (RAF1, BRAF, ARAF), MEKK1-4, DLK, and TAO2, regulate distinct branches of the MAPK pathway, including ERK, JNK, and p38 signaling. Dysregulation and mutation of MAP3Ks are implicated in cancer and inflammatory diseases, making them important targets for drug development. Due to their central role in cellular signaling, therapeutic targeting requires careful balancing to minimize toxicity while achieving efficacy.
