Mitogen-activated protein kinase kinase kinase 2 (MEKK2) mRNA is the transcript encoding the MEKK2 protein, a member of the MAP3K family (UniProt Q9Y2U5). MEKK2 is a key regulator of the JNK and ERK5 signaling pathways, which are vital for cell proliferation, survival, and the production of pro-inflammatory cytokines (PubMed: 10409743). In oncology, MEKK2 mRNA expression is often upregulated, promoting epithelial-mesenchymal transition (EMT) and metastasis in cancers such as breast and lung cancer (PubMed: 25605863). Targeting the mRNA via RNA interference (RNAi) or antisense oligonucleotides (ASOs) allows for the specific knockdown of MEKK2 protein levels, bypassing some of the challenges associated with small-molecule kinase inhibitors (PubMed: 30104359). Preclinical studies have demonstrated that silencing MEKK2 mRNA can inhibit tumor growth and reduce inflammatory responses in models of rheumatoid arthritis and intestinal inflammation (PubMed: 22431831). This target is particularly relevant for therapeutic interventions aiming to modulate complex signaling networks involved in chronic inflammation and malignancy.
