Mitogen-activated protein kinase kinase kinase 4 (MAP3K4) is a protein-coding enzyme that acts as a critical upstream regulator in the MAPK signaling cascade, activating kinases such as MAP2K4/MAP2K6 to trigger downstream JNK and p38 MAPK pathways in response to cellular stress, inflammation, and developmental stimuli. It is involved in a broad range of biological functions including apoptosis, autophagy, development, and stress response. MAP3K4 interacts with key regulatory proteins like GADD45, CDC42, RAC, and TRAF4, serving as an integrator of environmental and intracellular signals. Alterations in MAP3K4 expression or function have been associated with several diseases, notably with diverse roles in cancer, being implicated as both a tumor promoter and suppressor in different contexts. Potential targeting of MAP3K4 is under investigation for cancer therapy, but clinical application is challenged by its broad biological roles and the risk of unintended effects.
Drugs or inhibitors targeting MAP3K4 would generally act through kinase inhibition, blocking phosphorylation of downstream MAPK kinases (MAP2K4/MAP2K6), thereby attenuating JNK and p38 MAPK pathway signaling, influencing cell proliferation, stress response, and apoptosis.
