The TAK1-TAB1 complex is a central signaling module in the mitogen-activated protein kinase (MAPK) pathway. TAK1 (Transforming growth factor-β-activated kinase 1) is a serine/threonine kinase and a member of the MAPK kinase kinase (MAPKKK) family. It forms functional complexes with TAK1-binding proteins, primarily TAB1, but also TAB2 and TAB3. The assembly of these complexes is essential for the activation of downstream signaling pathways, including NF-κB and various MAPKs. It mediates inflammatory responses via NF‑κB pathway activation and regulates innate immune responses. As such it is considered a therapeutic target for diseases characterized by dysregulated inflammation or immunity such as autoimmune disorders or certain cancers.
