Overview

Mitogen-activated protein kinase p38 alpha (p38α or MAPK14) is a serine/threonine kinase belonging to the MAP kinase family. It plays a central role in cellular responses to stress, inflammation, cell differentiation, apoptosis, and immune responses. It is activated by dual phosphorylation by MKK3/MKK4 and regulates numerous transcription factors, influencing gene expression programs. Chronic activation is associated with inflammatory diseases, neurodegeneration, cardiac issues, and muscle diseases. While p38α inhibition has been explored for treating inflammatory and cardiac disorders, clinical outcomes have been mixed.
