This pathway regulates critical processes including cell growth, proliferation, differentiation, and survival (Wikipedia). Dysregulation of the MAPK pathway, often driven by oncogenic mutations in genes like BRAF or KRAS, is a major driver of tumorigenesis in various cancers, including melanoma and colorectal cancer (MDPI). Therapeutic strategies focus on small-molecule inhibitors targeting specific components like BRAF and MEK, which have significantly improved outcomes in mutation-positive patients (PubMed). However, clinical utility is frequently challenged by the rapid emergence of resistance through bypass signaling and paradoxical pathway activation in non-mutated cells (StatPearls).
