Mitoguardin 1 (MIGA1) is a mitochondrial outer-membrane protein crucial for the regulation of mitochondrial fusion, maintaining mitochondrial morphology, and establishing contacts between mitochondria and the endoplasmic reticulum (ERMCSs). MIGA1 promotes mitochondrial fusion through stabilization of mitochondrial phospholipase D and interacts with ER-localized proteins to regulate dynamic ER-mitochondria tethering. Genetic knockout models reveal its essential roles in female reproductive biology, including luteinization and progesterone production by ovarian granulosa cells, through control of mitochondrial structure and steroidogenic function. MIGA1 also connects mitochondria and autophagy, specifically mediating autophagosome-lysosome fusion and maintaining autophagic flux. Dysfunction of MIGA1 has been linked to neurodegeneration via impaired autophagy and mitochondrial function, and fusion events involving FAM73A (the gene encoding MIGA1) have been observed in cancer genomics databases, though no direct therapeutic targeting or pharmacological inhibitors are known for this protein. Thus, MIGA1 is primarily classified as a mitochondrial structural/regulatory protein with emerging importance in cell biology and disease, but not currently a direct drug target.
