Overview

Mitoguardin 2 (MIGA2) is a nuclear-encoded, mitochondrial outer membrane protein that plays a central role at ER-mitochondria contact sites where it mediates non-vesicular lipid transport—especially selective transfer of phosphatidylserine—from the ER to the mitochondria and potentially lipid droplets. It contains a lipid droplet targeting domain with a large hydrophobic pocket for lipid binding, and forms contacts via a phosphorylatable FFAT motif that interacts with ER-resident VAP proteins. MIGA2, along with its paralog MIGA1, also promotes mitochondrial fusion and is crucial for normal mitochondrial dynamics and function during oocyte meiosis, granulosa cell proliferation, and embryonic development in mammals. MIGA2 influences signaling pathways such as Hippo/YAP and AKT to facilitate cell proliferation, indicating its importance in reproductive endocrinology and potential roles in metabolic disease. Loss or dysfunction of MIGA2 is associated with metabolic defects and female subfertility in animal models, and may contribute to the molecular pathogenesis of disorders such as PCOS. No approved drugs or targeted therapeutics are currently known for MIGA2, and its pathway roles suggest a high level of cellular necessity and potential for safety concerns if targeted directly.
