Overview

Mitophagy is the selective degradation of mitochondria by autophagy, a process essential for maintaining cellular health and homeostasis. It specifically targets damaged or superfluous mitochondria, delivering them to lysosomes for breakdown and recycling. This process is evolutionarily conserved from yeast to humans and plays a critical role in mitochondrial quality control, energy metabolism adaptation, development (such as red blood cell maturation), and prevention of cellular degeneration. It involves recognition of damaged mitochondria, autophagosome formation, and fusion with lysosomes. Key pathways include PINK1-Parkin, BNIP3/NIX, and FUNDC1. Dysregulation of mitophagy contributes to various diseases, including neurodegeneration, cardiovascular disease, and metabolic disorders.
