Overview

Mitotic deacetylase-associated SANT domain protein (MIDEAS) is a nuclear protein that acts as a core component of the MiDAC (Mitotic deacetylase) histone deacetylase complex, also comprising HDAC1/2 and DNTTIP1[1][3][4]. The MiDAC complex regulates chromatin structure and gene expression by recruiting class I histone deacetylases to genomic loci, playing a key role in transcriptional repression and cell cycle-related processes; MIDEAS, through its ELM2-SANT domain, is critical for complex assembly and function, ensuring proper chromosome alignment during mitosis[1]. MIDEAS is essential for normal embryonic development, especially for proper heart morphogenesis and hematopoietic function, as demonstrated by mouse knockout models[1]. The MiDAC complex and its components, including MIDEAS, are implicated in several cancers and potentially in neurodegenerative disorders, although MIDEAS itself is not currently considered a direct therapeutic target nor known to be modulated by approved drugs[3][4][1].
