Overview

Mitotic kinesin Eg5, also known as Kinesin-5, KSP, or KIF11, is a member of the kinesin superfamily involved in spindle formation and maintenance during mitosis. As a homotetrameric bipolar motor protein, it crosslinks and slides apart anti-parallel microtubules, driving centrosome separation and maintaining spindle bipolarity. Eg5 moves toward the plus ends of microtubules using ATP hydrolysis. It is a validated target for antimitotic cancer therapies, and small-molecule inhibitors block its function, leading to mitotic arrest. Outside of cell division, Eg5 is involved in axonal growth regulation and may serve as a mobile link between ribosomes and microtubules.
