Overview

Mitogen-activated protein kinase kinase kinase 21 (MAP3K21) is a serine/threonine protein kinase and part of the MAP kinase kinase kinase (MAP3K) family. It acts as a negative regulator of TLR4 signaling and is involved in the regulation of signal transduction pathways, particularly those controlling immune responses. MAP3K21 is not known to activate the JNK, p38, or ERK2 MAPK pathways. It is predicted to have protein autophosphorylation and protein homodimerization activities, and is predominantly localized in the cytoplasm. The protein has been implicated in the pathogenesis of certain cancers (notably colorectal cancer, where it cooperates with activated RAS signaling to drive tumorigenesis) and as an immune marker in specific cancer subtypes[1][3][6][5]. No interacting drugs or significant safety concerns have been documented in the referenced sources.
