Target intelligence / Profile preview

Mixed lineage kinase domain-like protein (MLKL)

Target
MLKL
Molecular classification
Pseudokinase
01

Overview

MLKL is a pseudokinase that functions as the terminal effector of necroptosis. Upon activation by RIPK3-mediated phosphorylation, it oligomerizes and translocates to the plasma membrane, where it disrupts membrane integrity and triggers cell death. It has been implicated in various inflammatory, ischemic, and neurodegenerative diseases, as well as pediatric inflammatory bowel disease.
