MmpL3 is an essential inner membrane transporter in Mycobacterium tuberculosis responsible for exporting trehalose monomycolate (TMM). TMM is a precursor for mycolic acid synthesis, a crucial component of the mycobacterial cell wall. MmpL3 functions as a proton motive force-dependent antiporter, coupling inward proton movement with outward TMM transport. Inhibition of MmpL3 disrupts cell wall biosynthesis, leading to bacterial death, making it an attractive drug target for tuberculosis.
