Overview

MNX1 antisense RNA 2 (MNX1-AS2) is a long non-coding antisense RNA located at the *MNX1* gene locus on chromosome 7q36.3[1]. As an antisense transcript, it is distinct from the canonical MNX1 mRNA and is transcribed from a separate intergenic promoter. While much less studied than its paralog MNX1-AS1, MNX1-AS2 participates in the regulation of gene expression through epigenetic and transcriptional mechanisms, impacting biological processes such as cell cycle, chromatin remodeling, differentiation, and development[1]. Recent pan-cancer transcriptomic studies have demonstrated that MNX1-AS2, despite limited functional characterization, shows differential expression across various tumor types (notably gastrointestinal and testicular cancers) and can help distinguish between tumor subtypes and normal tissue with moderate accuracy. Its expression signature, particularly when combined with MNX1 and MNX1-AS1, holds promise as a biomarker for cancer diagnosis and possibly prognosis[1][2]. However, no therapeutic drugs directly target MNX1-AS2, and its precise molecular mechanisms remain largely unexplored.

04

Disease associations

Cancer (GI tract, testicular, pancreatic, esophageal, lung, endometrial, among others)Pre-eclampsia (rare, reported association)
05

Safety considerations

No direct safety concerns identified, as there are no drugs targeting this molecule clinically.General challenges include the specificity and delivery of lncRNA-targeting therapies, off-target effects, and interpretation of tissue-specific expression patterns for diagnostics

Biomarkers

MNX1-AS2 expression (alone or in combination with MNX1 and MNX1-AS1) is an emerging diagnostic and prognostic biomarker for distinguishing normal and tumor tissue, as well as specific tumor subtypes (adenocarcinoma vs. squamous, seminoma vs. non-seminoma), especially in gastrointestinal and testicular cancers
