MOB family member 4 (MOB4), also known as MOB-like protein phocein, is a member of the MOB kinase activator protein family, involved as a regulatory partner and scaffold in various signal transduction pathways, most notably the Hippo and STRIPAK complexes. It is a single domain adaptor protein (about 210–240 amino acids) that interacts with protein kinases to modulate phosphorylation events rather than directly catalyzing them. MOB4 antagonizes Hippo/MST1/2 kinase activation, thus modulating the output of the Hippo pathway which is crucial for cell proliferation, organ size control, and tumor suppression. MOB4 is expressed in several tissues and has been studied for its structural and regulatory roles, but currently is not considered a direct therapeutic target, nor is it targeted by approved drugs or used as a clinical biomarker. Disease associations are indirect, via pathways it modulates, especially in cancer biology[1][4].
