Overview

MOB kinase activator 1A (MOB1A) is an evolutionarily conserved regulatory protein and kinase activator essential for the Hippo signaling pathway. MOB1A acts as a coactivator of LATS1/2 kinases during the Hippo cascade, which regulates cell proliferation, apoptosis, and organ size[8][1][5]. Activation occurs through phosphorylation at specific threonine residues, triggering its binding to and stimulation of LATS1/2 activity and leading to downstream inhibition of YAP/TAZ nuclear translocation, thereby suppressing tumorigenic gene expression[1][5][8]. Loss or mutation of MOB1A leads to disrupted Hippo signaling, contributing to uncontrolled cell division and cancer formation, while its phosphorylation is associated with tumor suppression and proper cell cycle regulation[1][5][8]. The structural core of MOB1A is a zinc-stabilized four-helix bundle with a conserved, negatively charged surface, facilitating regulated electrostatic interactions with target kinases[4]. MOB1A is indispensable for the proper function of the Hippo pathway and thus is a critical indirect target in oncology and cell proliferation research.
