Overview

MOB kinase activator 2 (MOB2) is a member of the MOB (Mps One Binder) family of proteins, serving as a regulatory partner in kinase signaling pathways, particularly in the Hippo pathway and related NDR (nuclear Dbf2-related) kinase signaling[2][4]. It functions as a protein kinase activator, binding to and modulating the activity of serine/threonine kinases in the NDR/LATS family—most notably STK38 and STK38L[4]. MOB2 is localized in the cytosol, nucleolus, and nucleoplasm, and is implicated in processes such as actin cytoskeleton organization, neuron projection development, and regulation of protein phosphorylation[4]. Its binding helps activate or inhibit kinase partners depending on cellular context, and it can act both as a functional cofactor enhancing kinase function and, in some contexts, as a competitive antagonist of other MOB family members' interactions with those kinases[2]. No direct pharmacologic agents targeting MOB2 are known, nor is it a common direct biomarker or drug safety concern. Diseases linked to its genetic locus include Carpal Tunnel Syndrome 1 and mild Mononeuropathy of the median nerve[4].
