Monkeypox virus antigen H3 (H3L protein) is a major envelope glycoprotein on the surface of the monkeypox virus. It mediates viral adhesion and entry into host cells by binding to cell-surface heparan sulfate. H3 is a major antigenic target recognized by both humoral and cellular immune responses, eliciting strong antibody responses and T-cell interferon responses. Monoclonal antibodies against H3 can neutralize MPXV in vitro and protect animals from lethal poxvirus challenge. It is used in rapid diagnostic assays and is a vaccine candidate due to its strong immunogenicity and conservation across orthopoxviruses.
