C-C motif chemokine ligand 8 (CCL8) is a small secreted cytokine of the CC chemokine family, also known as monocyte chemoattractant protein 2 (MCP-2). It is encoded by the CCL8 gene on chromosome 17 in humans and functions to attract and activate various immune cells—including monocytes, T cells, NK cells, eosinophils, and basophils—by binding to several chemokine receptors (notably CCR1, CCR2B, CCR3, and CCR5)[3]. CCL8 has roles in mediating inflammatory responses, recruiting macrophages, and modulating fibrosis. Elevated CCL8 expression is associated with inflammatory diseases, several cancers, and fibrotic conditions. It is considered a therapeutic target, as neutralizing antibodies against CCL8 have shown efficacy in reducing inflammation and fibrosis in experimental models[2][4][3]. CCL8 is also a prognostic biomarker in some cancers, including diffuse large B-cell lymphoma[4][6].
Neutralization of CCL8 (by antibodies) blocks its chemotactic signaling via CCR receptors, which reduces immune cell recruitment and associated inflammation or fibrosis[2]
