Monocyte to macrophage differentiation factor (MMD, also known as PAQR11) is a member of the progestin and adipoQ receptor (PAQR) family, structurally characterized by seven predicted transmembrane domains but not functioning as a classical G protein–coupled receptor. Localized primarily to the Golgi apparatus, MMD acts as a scaffold protein crucial for monocyte to macrophage differentiation and regulates cellular survival by promoting anti-apoptotic mechanisms via the MAPK/ERK pathway, and by facilitating Ras signaling in concert with RasGRP1. MMD is upregulated during pathological conditions such as rheumatoid arthritis and certain cancers, functions in cell proliferation, immune modulation, and enhances sensitivity to ferroptosis by altering phospholipid metabolism. Its disease associations and regulatory roles in signaling and cell fate decisions make it a candidate therapeutic target in inflammation, cancer, and diseases involving disordered macrophage function or ferroptosis[1][2][3][5].
Not established for clinical drugs; experimental modulation includes RNA interference/knockdown, impacting cell proliferation, Ras/ERK signaling, and ferroptosis by influencing lipid remodeling and apoptosis[5][1][2].
