Overview

Monovalent cations are positively charged ions with a valence of one, most notably sodium (Na+), potassium (K+), and hydrogen (H+) (NIH, 2016). They play a fundamental role in biological systems by establishing electrochemical gradients across cell membranes, which are essential for nutrient transport, pH regulation, and the generation of action potentials in neurons and muscle cells (StatPearls, 2023). In addition to their role as charge carriers, certain monovalent cations act as allosteric modulators or essential cofactors for enzymes like thrombin and pyruvate kinase (PNAS, 2011; NIH, 2012). While the ions themselves are not therapeutic targets in the traditional sense, the proteins that regulate their movement—such as ion channels, transporters, and the Na+/K+-ATPase pump—are major targets for drugs like diuretics, anti-arrhythmics, and digitalis glycosides (UniProt, 2024). Furthermore, lithium, a monovalent cation itself, is used as a primary treatment for bipolar disorder, though its exact molecular targets are diverse (PubMed, 2021). Dysregulation of these ions is associated with critical conditions such as hypertension, heart failure, and electrolyte imbalances (NIH, 2023).
