Overview

Montanide refers to a family of water-in-oil emulsion adjuvants, most notably Montanide ISA 51 and Montanide ISA 720, used to enhance immune responses in vaccine formulations. It acts primarily by stimulating the immune system to generate stronger and more durable responses against co-administered antigens through a depot effect, enhanced antigen presentation, and activation of innate immunity, promoting both humoral and cellular immune responses.

Mechanism of action

Montanide's adjuvant effect is based on several immunological mechanisms: - Depot Effect: The water-in-oil emulsion forms a depot at the injection site, allowing for sustained release of antigen over time. This prolonged exposure increases the duration during which antigen-presenting cells (APCs) can interact with the antigen. - Enhanced Antigen Presentation: By facilitating slow release and local concentration of antigens, Montanide increases uptake by APCs such as dendritic cells. These APCs process the antigen and present it on MHC class II molecules to T cells, initiating adaptive immunity. - Activation of Innate Immunity: The emulsion induces local inflammation at the injection site, recruiting immune cells including macrophages, dendritic cells, B lymphocytes, and T lymphocytes. This leads to upregulation of cytokines/chemokines that further stimulate both innate and adaptive arms. - Promotion of Humoral Response: Studies show that Montanide-adjuvanted vaccines induce higher titers of antibodies with greater avidity for their target antigens compared to some other adjuvants. In particular, Montanide ISA 720 has been shown in animal models to promote differentiation toward plasma cell lineages through gene signatures associated with heme biosynthesis in B lymphocytes. - Mixed Th1/Th2 Cytokine Profile: Immunization with Montanide-based formulations often results in a mixed Th1/Th2 cytokine response profile—supporting both cellular (T cell-mediated) and humoral (antibody-mediated) immunity.
