Overview

The λ2^315^ tumor idiotype neoantigen is a specific peptide derived from the variable region of the lambda-2 light chain of the MOPC315 myeloma protein (Bogen et al., 1986). In the MOPC315 mouse model, this idiotype contains somatic mutations that create a unique neoantigenic epitope recognized by the immune system (Lauritzsen et al., 1994). When processed and presented on MHC class II molecules (specifically I-E^d), this peptide is recognized by idiotype-specific CD4+ T cells, which can orchestrate a potent anti-tumor response (Corthay et al., 2005). This recognition triggers the activation of macrophages and other effector cells, leading to the rejection of myeloma cells even if they do not express MHC class II themselves (Bekri et al., 2022). It serves as a foundational model for studying how the immune system can be primed to recognize tumor-specific antigens and the critical role of CD4+ T cells in anti-tumor immunity. Therapeutic strategies targeting this neoantigen include idiotype-based DNA or peptide vaccines and the adoptive cell transfer of TCR-engineered T cells (Lauritzsen et al., 1993).
