Overview

MORC family CW-type zinc finger protein 1 (MORC1) is a nuclear protein characterized by a GHKL-type ATPase domain, a CW-type zinc finger, and several coiled-coil domains, and is considered an epigenetic regulator. MORC1 is critical for spermatogenesis and functions in the silencing of transposable elements in male germ cells by facilitating chromatin methylation and compaction, specifically the establishment of H3K9me3-marked heterochromatin on transposon-rich genomic regions. While primarily studied in the testis, it is also expressed in the brain, with evidence suggesting a role in neurodevelopment and mood regulation—possibly acting as a transcription factor that mediates susceptibility to early life stress and depression via epigenetic mechanisms. MORC1 loss-of-function is associated with male infertility and has been linked to neuropsychiatric and neuromuscular disorders. Its expression and DNA methylation state are under investigation as potential biomarkers for certain mood disorders and developmental conditions. No drugs are currently known to pharmacologically target MORC1, and disruption of its function could raise significant safety concerns for fertility and brain development.
