MORN repeat-containing protein 3 (MORN3) is a protein encoded by the MORN3 gene. It contains MORN (Membrane Occupation and Recognition Nexus) repeats, a domain type found across many species and assumed to mediate protein-protein or protein-lipid interactions[1]. In humans, MORN3 is recognized for its role in assembling a suppression complex that tethers SIRT1 and MDM2 to regulate modifications of the tumor suppressor p53/TP53. This regulation includes both deacetylation (by SIRT1, leading to functional inactivation) and ubiquitin-dependent degradation (by MDM2), thus promoting proliferative and cell survival behaviors[3]. MORN3 is located in the nucleus and is involved in the negative regulation of signal transduction by p53 class mediator[3]. There may be a role for MORN3 in spermatogenesis, but its functions remain relatively poorly characterized. No drugs or small molecules are currently known to directly target MORN3, and it is not established as a therapeutic target.
