MORN repeat-containing protein 5 (MORN5) is a cytoplasmic protein characterized by the presence of multiple MORN (Membrane Occupation and Recognition Nexus) repeats, which are protein-protein interaction modules found across eukaryotes and prokaryotes[1][3]. MORN5 plays a role in embryonic development, particularly in craniofacial structures, where it is expressed in a highly specific spatial and temporal pattern during facial morphogenesis in vertebrates, including chick and human embryos[2]. It is regulated by and required for BMP (Bone Morphogenetic Protein) signaling, and modulates the expression of genes in both BMP and TGFβ (Transforming Growth Factor beta) pathways[2]. Variants in MORN5 have been associated with increased risk of non-syndromic cleft lip with or without cleft palate in humans, suggesting a clinically relevant developmental function[2]. To date, there is no evidence that MORN5 acts as a therapeutic target, receptor, enzyme, or has direct pharmacological modulation, and its biochemical functions remain incompletely defined[3][1][2].
