Overview

Mortality factor 4-like protein 1-associated protein (MRGBP) is a core component of the human NuA4 histone acetyltransferase (HAT) complex, which is responsible for the acetylation of histone H4 and H2A (UniProt Q9NVW5) [1]. This complex plays a fundamental role in transcriptional activation, DNA repair, and cell cycle progression by modifying chromatin structure (GeneCards) [2]. MRGBP specifically interacts with MORF4L1 (MRG15) to stabilize the complex and facilitate its recruitment to specific genomic loci (PubMed: 15145945) [3]. In oncology, MRGBP is recognized as a potential therapeutic target because its overexpression is associated with the progression of various cancers, including colorectal and prostate cancer, where it promotes cell proliferation and metastasis (PubMed: 30212440) [4]. Although no clinical-stage drugs currently target MRGBP, research suggests that disrupting its interaction within the NuA4 complex could inhibit tumor growth and sensitize cells to DNA-damaging agents (PubMed: 25605116) [5].
