Overview

The mosquito larval midgut is the central digestive organ of the mosquito larva, serving as the primary site for nutrient processing, absorption, and ion regulation [1]. It is composed of a single layer of epithelial cells protected by a semi-permeable peritrophic matrix, which acts as a critical interface between the insect and its aquatic environment [2]. In the field of vector control, the midgut is the principal target for biological insecticides such as Bacillus thuringiensis israelensis (Bti) and Lysinibacillus sphaericus [3]. These agents produce protoxins that are solubilized in the highly alkaline environment of the midgut and activated by larval proteases to bind specific receptors, such as cadherins and aminopeptidases [4]. This binding leads to pore formation in the epithelial cell membranes, causing osmotic lysis, midgut paralysis, and the eventual death of the larva [5]. By targeting the larval midgut, these strategies effectively reduce the population of adult mosquitoes capable of transmitting diseases like malaria, dengue, and Zika virus [6].
