Overview

SMAD7 is a key intracellular antagonist of the transforming growth factor-beta (TGF-β) signaling pathway. It acts as a negative feedback regulator by binding to activated TGF-β type I receptors, thereby blocking the phosphorylation and activation of receptor-regulated Smads (R-Smads). SMAD7 also inhibits signaling from activin and bone morphogenetic protein (BMP) pathways. Its expression is induced by TGF‑β, EGF, IFNγ, and TNFα. Mutations or altered expression levels of SMAD7 are associated with several cancers, and it also modulates inflammatory processes.

Mechanism of action

Inhibits TGF-β, activin, and BMP signaling by binding to type I receptors and preventing R-Smad phosphorylation/activation. Blocks IL-1R/TLR complex and reduces NF-kB activity by binding Pellino-1.
