Smad7 mRNA encodes mothers against decapentaplegic homolog 7, a key intracellular inhibitory modulator of the TGF-β family signaling pathways through negative feedback on receptor-mediated Smad2/3 activation. Smad7 mRNA is the immediate target for antisense therapies (like Mongersen), and it plays major roles in controlling cellular proliferation, inflammation, apoptosis, and tissue homeostasis.
