Motile sperm domain-containing protein 1 is a membrane-anchored protein containing a major sperm protein (MSP) domain and two transmembrane regions. It is widely expressed, especially in mesenchymal tissues, and localizes mainly to the Golgi apparatus and other cellular membranes. MOSPD1 is involved in cell signaling, differentiation—particularly of mesenchymal stem cells—and cytoskeletal organization, affecting cell shape and motility[1][4][5]. Recent evidence implicates MOSPD1 as a tumor-promoting factor in breast cancer, correlating with poor clinical outcomes and immune evasion, and its inhibition sensitizes tumor cells to immune checkpoint therapy. Its function is dependent on intracellular localization and regulation of mesenchymal versus epithelial gene expression, indicating a role in developmental transitions and possibly other disease processes[1][2][4]. There are currently no approved drugs specifically targeting MOSPD1, but its biomarker and therapeutic target potential in cancer is being investigated[2].
For therapies targeting MOSPD1 (preclinical context): Suppression of MOSPD1 inhibits tumor growth and increases apoptosis. Enhances response to immune checkpoint inhibitors (e.g., anti-PD-L1 therapy).
